MODELLING AND OPTIMIZING EXTRACTION SOLVENT MIXTURE FOR BIOACTIVE PHYTOCHEMICAL EXTRACT PRODUCTION FROM HYPTIS SUAVEOLENS (L) LEAVES

ABSTRACT

This study was done to investigate the effects of solvent mixtures on the extraction of phenolic-rich bioactive compounds from Hyptis suaveolens leaves. In this paper, various proportions of water and ethanol between 0 and 100%, were investigated to obtain the optimal binary solvent mixture proportional composition. From results obtained, the factors of solvent ratios in the mixture were found to significantly affect the extraction outcomes. This was observed in the results of the dependent variables, which were used for the study. Responses of total phenolic content (TPC), total flavonoid content (TFC) and antioxidant activities of 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging capacity were used for evaluation in the study. From the results obtained in the study, 25% ethanol and 75% water were found as the optimal proportions of the binary solvent mixture, while TPC, TFC and DPPH of 13.63mg GAE/g d.w., 9.60mg QE/g d.w. and 2.42 µg/mL respectively, were obtained as responses of the phenolic-rich bioactive compounds in the extract obtained at the optimal binary mixture composition. From the results of Fourier transformed infra-red spectroscopy (FT-IR) characterization of the extract strong peaks of functional groups (O-H, CH3, C-O, and C-C), which indicated the presence of hydroxyl, lipids, and alkanes were revealed. The result also showed medium and weak bands, highlighting the presence of other groups, such as secondary alcohol, alkene and amino acid. The type of bioactive compounds found in the extract of Hyptis suaveolens leaves indicated that the plant has potential for medicinal application.
